What is Kisspeptin?

Kisspeptin-10 is a ten-amino-acid fragment of kisspeptin, the peptide encoded by the KISS1 gene, and the shortest fragment that retains full activity at its receptor, KISS1R (also called GPR54). It is sold strictly as a research chemical: it is not approved by the FDA or any other regulator for human use, and nothing on this page is medical advice.

Its evidence base is unusual for this catalogue. Alongside the rodent, primate and cell-culture work that established the receptor, there is human genetic evidence and a body of supervised clinical infusion research. That human work was done in academic trials with pharmaceutical-grade material; no kisspeptin product is approved anywhere, and research-grade material is not that material.

Mechanism of Action

The mechanisms below come from receptor pharmacology in cell lines, from rodent and non-human primate studies, and — unusually for a research peptide — from human clinical genetics and supervised infusion research. Each is labelled with the setting it came from.

Receptor target. Kisspeptin-10 is the C-terminal decapeptide of the KISS1 gene product and acts as an agonist at KISS1R (GPR54), a Gq/11-coupled receptor. In transfected cell lines, binding activates phospholipase C, releases inositol trisphosphate and diacylglycerol, mobilises intracellular calcium and phosphorylates ERK1/2. This was the pharmacology that identified the peptide as the receptor's endogenous ligand.

Upstream control of GnRH. Kisspeptin-expressing neurons in the hypothalamus project to GnRH neurons, which carry the receptor. In rats, mice, sheep and monkeys, kisspeptin-10 given centrally or peripherally triggers GnRH release and, downstream of it, luteinising hormone and follicle-stimulating hormone. This literature describes the peptide as acting on the GnRH pulse generator rather than on the pituitary directly, and reports that continuous exposure desensitises the response in primates while intermittent exposure does not — the pattern of exposure changes the result.

Human genetic evidence. Loss-of-function mutations in GPR54 cause normosmic idiopathic hypogonadotropic hypogonadism, in which puberty does not begin, and activating mutations have been reported in central precocious puberty. This is the strongest human evidence that the pathway is required for normal reproductive development, and it comes from clinical genetics — from people who inherited a broken receptor — not from giving anyone the peptide.

Human infusion studies, all investigational. Academic groups have infused kisspeptin-10 and the longer kisspeptin-54 into volunteers under clinical supervision and measured gonadotropin responses, and kisspeptin-54 has been trialled as an oocyte maturation trigger in IVF. Kisspeptin-10 is cleared from circulation within minutes, considerably faster than kisspeptin-54, so results with one fragment are a poor guide to the other. This work is investigational research with pharmaceutical-grade material under monitoring; it is not evidence about a research-grade vial and contains nothing that functions as a protocol.

The original context was cancer biology. KISS1 was first identified as a metastasis-suppressor gene in human melanoma cell lines, which is why the peptide was originally named metastin. That anti-metastatic activity was characterised in cell culture and in mouse models and has not produced an approved therapy.

None of these findings establishes safety or efficacy outside a supervised trial. No kisspeptin product is approved for any medical use in any country; the peptide has been given to people only inside clinical research, and the research-grade material sold by these vendors is not that trial material and is not a medicine.
