What is CJC-IPA?

CJC-1295 (no DAC) + Ipamorelin is a blend of two compounds in one vial, not a single peptide. CJC-1295 without DAC — also sold as modified GRF (1-29) — is a 29-amino-acid analogue of growth hormone-releasing hormone; ipamorelin is a synthetic pentapeptide acting at a different receptor. It is sold strictly as a research chemical: not approved by the FDA or any other regulator for human use, and nothing on this page is medical advice.

The two components have been studied separately — mostly in rodents and pigs, plus early-phase human pharmacology measuring blood hormone levels. The blend itself has no dedicated study base: no trial has tested this combination, the case for pairing them extrapolated from older GHRH-plus-secretagogue work.

Mechanism of Action

The blend has no mechanism of its own. What follows is the mechanism described for each component in the literature, plus the reasoning behind pairing them — which comes from work on other compound pairs, not on this one.

Two receptors, not one. The two components reach the same cells by different doors. CJC-1295 without DAC acts at the growth hormone-releasing hormone receptor on pituitary somatotrophs; ipamorelin acts at the growth hormone secretagogue receptor (GHS-R1a), the receptor ghrelin binds. Both are described in animal work as converging on growth hormone release from the anterior pituitary.

The GHRH component. CJC-1295 without DAC is the 1-29 fragment of growth hormone-releasing hormone carrying four amino acid substitutions intended to slow enzymatic breakdown, dipeptidyl peptidase-4 in particular. In rats, analogues of this family activate the pituitary GHRH receptor and raise circulating growth hormone. An important distinction for reading the literature: the version tested in healthy adults was CJC-1295 with DAC, an albumin-binding conjugate that persists for days. The no-DAC form lacks that conjugate and is described as short-lived, so the human data on the DAC form does not transfer to it.

The ipamorelin component. Ipamorelin is a synthetic pentapeptide characterised in rat and swine models as a selective growth hormone secretagogue — in those models growth hormone release occurred with little change in ACTH, cortisol or prolactin, which is what distinguished it from earlier peptides such as GHRP-6. It was carried into early-phase human pharmacology, where the measured endpoint was hormone concentration in blood, and into later trials for postoperative ileus that did not lead to approval for any indication.

Why the two are combined. In human studies from the 1980s and 1990s, a GHRH given together with a growth hormone-releasing peptide produced more growth hormone release than either produced alone; the pathways are described as separate, the GHRH side signalling through cAMP and the secretagogue side acting through a calcium-dependent route while lowering somatostatin tone. That synergy was demonstrated for other GHRH and GHRP pairings. It is the stated rationale for this blend, not a finding about it.

What was actually measured. Across nearly all of this work the outcome recorded is a hormone concentration — growth hormone pulses, or IGF-1 downstream — rather than a clinical result. There is no published pharmacokinetic or safety data for the combination as vendors sell it, and the ratio of the two components varies between products, so even the component-level findings above cannot be read across to a particular vial.
