What is BPC-157?

BPC-157 (Body Protection Compound-157) is a pentadecapeptide composed of 15 amino acids. It is a partial sequence of the body protection compound (BPC) that is discovered in and isolated from human gastric juice. Experimentally it has been demonstrated to accelerate the healing of many different wounds, including tendon-to-bone healing and damaged ligaments. BPC-157 has shown particular promise in healing soft tissue injuries and has been studied for its potential therapeutic effects on various organ systems.

Mechanism of Action

The mechanisms proposed for BPC-157 come from rodent and cell-culture work, not from human pharmacology, and several remain hypotheses rather than settled findings.

Angiogenesis and vascular signalling. In rat tendon and muscle injury models, treated tissue shows increased expression of vascular endothelial growth factor receptor 2 (VEGFR2) and faster formation of new blood vessels around the injury. The authors propose this as the route by which the peptide accelerates the early phase of healing.

Nitric oxide system. A recurring finding is interaction with the NO pathway: in the same animal models BPC-157 is reported to counter both the effects of NO-synthase inhibition (L-NAME) and of NO over-production (L-arginine). Researchers describe this as a modulating effect rather than a simply stimulating one.

Tendon fibroblast signalling. In cultured tendon fibroblasts, BPC-157 has been reported to increase expression of the growth hormone receptor and to activate the FAK–paxillin pathway, which is associated with cell migration and spreading. This is one proposed explanation for the tendon-to-bone healing results seen in rat Achilles models.

Gastrointestinal cytoprotection. The peptide was originally studied for gastric and intestinal lesions, where it is reported to help maintain mucosal integrity and to interact with the dopaminergic and serotonergic systems along the gut–brain axis.

Stability. Unlike most peptides, BPC-157 is reported to remain stable in human gastric juice for more than 24 hours, which is why animal studies have used oral as well as injected routes.

None of these findings establishes safety or efficacy in humans. The compound is not approved for any medical use.
